Redox active metalloenzymes catalyse a range of biochemical processes essential for life. However, due to their complex reaction mechanisms, and often, their poor optical signals, detailed mechanistic understandings of them are limited. Here, we develop a cryoreduction approach coupled to electron paramagnetic resonance measurements to study electron transfer between the copper centers in the copper nitrite reductase (CuNiR) family of enzymes. Unlike alternative methods used to study electron transfer reactions, the cryoreduction approach presented here allows observation of the redox state of both metal centers, a direct read‐out of electron transfer, determines the presence of the substrate/product in the active site and shows the importance of protein motion in inter‐copper electron transfer catalyzed by CuNiRs. Cryoreduction‐EPR is broadly applicable for the study of electron transfer in other redox enzymes and paves the way to explore transient states in multiple redox‐center containing proteins (homo and hetero metal ions). 相似文献

Anopheles stephensi is a mosquito vector of malaria, which is still considered a relevant public health problem due to increasing outdoor transmission, growing resistance to insecticides used to target vectors, and antiplasmodial drugs as well. Thus, there is a vital need to explore novel sources of effective compounds. In this study, the hydrothermal method was used for the synthesis of bismuth oxyiodide (BiOI) nanoflakes. Furthermore, the toxicity of BiOI nanoflakes was evaluated for the first time on A. stephensi, as well as in vivo against the malaria parasite Plasmodium berghei. The synthesis of BiOI nanoflakes was confirmed by various characterization techniques, including X-ray diffraction, Fourier transform-infrared spectroscopy, field emission scanning electron microscopy and transmission electron microscopy (HR-TEM). LC50 of BiOI nanoflakes on A. stephensi were 2.263 ppm (larva I), 3.414 ppm (II), 4.956 ppm (III), 6.983 ppm (IV) and 8.605 ppm (pupae). In vivo antiplasmodial experiments conducted on P. berghei infecting albino mice showed 27.2% of chemosuppression after 4 days of treatment with 300 mg/kg/day of BiOI, a lower performance if compared to chloroquine. Overall, our results suggested that hydrothermal synthesis of BiOI nanoflakes may be considered to develop newer and safer tools for malaria vector control. 相似文献

We have developed a flow cell that allows culturing adherent cells as well as suspended cells in a stable, homogeneous, and low-shear force environment. The device features continuous medium supply and waste exchange. In this paper, a simple and fast protocol for device design, fabrication, and assembly (sealing) based on a poly(dimethylsiloxane) (PMDS)/glass slide hybrid structure is described. The cell culture system performance was monitored, and the effective shear force inside the culture well was also determined. By manipulating the device dimensions and volumetric flow rate, shear stress was controlled during experiments. Cell adhesion, growth, proliferation, and death over long-term culture periods were observed by microscopy. The growth of both endothelial and suspension cells in this device exhibited comparable characteristics to those of traditional approaches. The low-shear culture device significantly reduced shear stress encountered in microfluidic systems, allowing both adherent and suspended cells to be grown in a simple device. 相似文献

Synthesis of transparent and crack-free monoliths of silica aerogel by sub-critical drying technique is reported in the present
article. Silane ageing with 50% tetraethylorthosilicate:ethanol followed by solvent exchange using ethanol was adopted. The
effect of heat-treatment on the textural and physical characteristics of silica aerogel was evaluated. The chosen composition
resulted in a high surface area silica aerogel of 1,000 m2 g−1 and a pore volume of 1.4 cm3 g−1 at room temperature. The aerogel heat-treated at 900 °C possessed a surface area of 450 m2 g−1 with a pore volume of 0.4 cm3 g−1. The decrease in surface area and pore volume was associated with the sintering process. The present technique seems advantageous
in preserving the high surface area of the material at high temperatures. The XRD studies showed that the amorphous nature

The effect of alkali treatment and fiber length on the wear performance of the Palmyra palm leaf stalk fiber (PPLSF)–polyester composites and the possibilities for using PPLSF in wear resistance applications were explored at different speeds and normal loads for constant sliding distance using pin on disk wear tester as per the ASTM G99 standard. Unsaturated polyester was used as matrix, and composites were prepared by molding in an open mold and pouring the resin. It was observed that wear loss and coefficient of friction reduced due to reinforcement of PPLSF. Reinforcing alkali-treated PPLSF in the matrix has further reduced the wear loss and coefficient of friction. At high speed and high load condition considered in this study shows that the wear loss and coefficient of friction were reduced by 64 and 22%, respectively, for alkali-treated fiber composites compared with untreated fiber composites. The effect of fiber length on the wear performance was also evaluated and optimal set of parameters that would result in minimum wear loss and coefficient of friction was determined by design of experimental method using Taguchi’s orthogonal array. The surface morphology of the composites after wear tests was examined using scanning electron microscopy to analyze the mechanism of wear. 相似文献

Atomic force microscopy (AFM) has been successfully used to study the activation energy for evaporation of pentaerythritol tetranitrate (PETN) nanoislands formed by spin coating. These islands are annealed isothermally in the temperature range of 30-70 degrees C for a given time and are scanned with AFM in contact mode at room temperature. The volume of these islands does not change significantly up to about 35-40 degrees C indicating that sublimation is not significant below 40 degrees C. Above 40 degrees C, the islands start shrinking, and the rate of weight loss is analyzed as a function of temperature. The activation energy of evaporation using AFM was found to be similar to that for bulk PETN crystals using thermogravimetric analysis (TGA) at higher temperatures (110-135 degrees C). These results demonstrate that AFM is a useful tool to measure thermodynamic properties with a nanoscale probe. 相似文献
